Deficiency of Th17 cells in hyper IgE syndrome due to mutations in
STAT3. J Exp Med 2008; 205(7): 1551-7.
"We knew something was very wrong and at two years, fortunately, he was offered a DNA test and it was then we found out he had a
STAT3 gene mutation.
Because
Stat3 regulates many cellular processes, the scientists combed through genes expressed during muscle growth to find additional proteins regulated by
Stat3 that might serve as more specific targets.
Antibodies specific to p-STAT3,
STAT3, COX-2, MMP-2, MMP-9 were purchased from Cell Signaling Technology (Shanghai, China) while antibodies specific to E-cadherin, N-cadherin and Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) were purchased from Proteintech (Wuhan, Hubei, China).
KCs were seeded into sterilized 96-well plastic plates made of polystyrene and transfected with 100 nmol/L
STAT3 siRNA (LUS group) or siRNA-NC carried by Lipofectamine 3000 associated with SonoVue microbubbles and then irradiated by the probe of ultrasonic transfer apparatus through ultrasound couplant (Yalian, China) from the bottom of the plates.
Additionally, IL-17 can directly stimulate collagen production in HSCs through
STAT3 activation [31], and
STAT3 is also suggested to play a protective role in the liver by enhancing hepatocyte regeneration [32].
Consistent with previous studies, we found that infusion of MSCs activated
STAT3 signaling, exhibiting increased expression of p-STAT3 at 24 h (Figure 5(c)).
With a high suspicion of AD-HIES, mutation analysis of
STAT3 gene was sent which detected a novel pathogenic mutation, C.1388 T> A (pVal463Glu) at DNA-binding domain of
STAT3 gene.
We identified the correct LC concentrations as the lower concentrations that activated
STAT3 signaling.
STAT3 signaling regulates various biological processes, such as cell growth, survival, differentiation, and apoptosis [7].
STAT3 can be activated through activating with gp130.
