podophyllotoxins

podophyllotoxins

A range of powerful substances derived from the mandrake root (Podophyllum peltatum ). Podophyllotoxins act as TOPOISOMERASE inhibitors causing breaks in DNA strands. Ironically in view of John Donne's famous reference to mandrake, podophillotoxins are also used locally to treat genital WARTS in both sexes. Brand names of preparations for the latter purpose are Condyline and Warticon.

The podophyllotoxins (etoposide and tenisopide) are derived from the May apple plant (Podophyllum peltatum).
Podophyllotoxins and camptothecan analogs are also effective as Topoisomerase inhibitors.
Canel says the mayapple adds a glucose molecule to podophyllotoxins so the compound can be safely stored until the plant is attacked.
This allowed the release of large amounts of stored podophyllotoxins," says Canel.
While, etoposide (Eto) is a semi synthetic derivative of the podophyllotoxins, which inhibits DNA synthesis by inhibiting DNA topoisomerase II which is cell cycle dependent and phase specific, affecting mainly the S and G2 phases (19).
They are semisynthetic derivatives of podophyllotoxin, a lignan derived from American mandrake (Podophyllum peltatum L).
To begin with, the compounds from higher plants include Berbamine, Butulinic acid, Camptothecin, Cucurbitacin, Ellipticine, Flavopiridol, Homoharringtonine, Silvestrol, Berberine, Daphnoretin and Podophyllotoxin.
Beside well-established anticancer drugs such as etoposide and temposide, highly cytotoxic phytochemicals such as podophyllotoxin were included in this panel of compounds.
These methods have yielded important discoveries including vincristine, vinblastine, the podophyllotoxin derivatives, lO-hydroxy-camptothecin and Taxol (Suffness and Douors, 1979; Wani et al.
