metal shadowing

A technique used to obtain information about the shape of purified viruses, fibres, enzymes and subcellular particles.

Method
A thin layer of evaporated metal (e.g., platinum) is sprayed at an angle to the biological specimen; an acid bath then dissolves the biological material leaving the metal replica, which is then examined by transmission electron microscopy.
