conformational epitope

conformational epitope

Adjacent amino acid strings at different points in a folded protein, making a complex site to which an antibody can bind. Conformational epitopes are less stable than LINEAR EPITOPES, a fact that explains why some allergies are outgrown in time. Allergies, such as peanut allergy, in which the antibody (IgE) binds to a linear epitope tend to persist.

Because SmBB' and U1-nRNP share a cross-reactive epitope, the SmD1 protein is likely the most important polypeptide in the Sm antigen.[sup][9] Among SmD1 peptides, the SmD1-amino-acid 83-119 peptide (SmD1[sub]83-119), which was discovered through epitope mapping using 13-mer peptides overlapping by ten amino acids and sera from adult SLE patients, was described as a major epitope targeted by autoantibodies in adult SLE sera.[sup][10] Researchers further revealed that the polypeptide was a conformational epitope that was not accessible in the full-size SmD1 protein.
An aromatic patch is formed in the central part of the protein by 2 tryptophan and 1 tyrosine residues encircled by 4 glutamate side chains forming a carbohydrate-combining site constituting a conformational epitope [110].
Conformational epitope comprises amino acids that are close in space in the folded molecule, despite being noncontiguous in the amino acid sequence.
(10) Although potential binding sites have been identified through the knowledge obtained from the crystal structure of AQP-4, it is not clear yet whether these antibodies recognize a linear or a conformational epitope. Thus the simple approach, comparing the amino acid sequence of potential binding sites between humans and rodents, is not possible.
