ag·gre·can

(ag're-kan),
Candidate gene for otosclerosis located at 15q25 to q26.

Aggrecan

A proteoglycan expressed in variant forms through alternative splicing which, with type II collagen, is a major component of articular cartilage and other cartilaginous tissues. Aggrecan is broken down by the ADAMTSs (A Disintegrin And Metalloprotease with ThromboSpondin) protein family, ADAMTS4 (aggrecanase-1) and ADAMTS5 (aggrecanase-2). ADAMTS1 processes aggrecan and forms a covalent-binding complex with alpha2-macroglobulin which is dependent on the zinc-binding site.

ag·gre·can

(ag'rĕ-kan)
Candidate gene for otosclerosis located at 15q25-q26.

aggrecan

An extracellular PROTEOGLYCAN that forms exceptionally large aggregates giving cartilage its particular gel-like properties and resistance to deformation.

aggrecan

the shortened name for the large aggregating chondroitin sulphate proteoglycan.

SB-061, which will be administered as an intra-articular injection, is designed to prevent the degradation of cartilage by mimicking the protective effect of the proteoglycan aggrecan.
The breakdown of aggrecan, in particular, which is included in the extracellular matrix, is critical in articular damage.
Researchers in Grodzinsky's lab are now working to identify possible drugs that might halt the loss of aggrecan, as well as designing tissue scaffolds that could be implanted into patients who need cartilage-replacement surgery.

Explant and monolayer cell culture data differed in the amount of gene activity observed, especially for aggrecan and Collagen Type X, but MMP-13 was reduced by vitamin C at both normoxic and hypoxic levels.
FGF-2 activates a compound called Sox9 in the stem cell, which in turn switches on the production of two main components of cartilage, type 2 collagen and aggrecan, Li says.
In these cells, it is noteworthy that aggrecan and type X collagen are constitutively expressed.
