UHRF1

UHRF1

A gene on chromosome 19q13.3 that encodes a member of a subfamily of RING-finger type E3 ubiquitin ligases, which binds to specific DNA sequences and recruits a histone deacetylase to regulate gene expression. Its expression peaks at late G1 phase, and plays a major role in the G1/S transition by regulating topoisomerase II-alpha and retinoblastoma gene expression; it also functions in the p53-dependent DNA damage checkpoint.

Additionally, the expression of UHRF1, the protein that recruits DNMT1 to hemimethylated DNA sites, was significantly reduced in a dose-dependent manner after PEP exposure.
Hypomethylation may also be mediated by decreased levels of UHRF1, which specifically interacts with DNA methyltransferases and hemimethylated sites on DNA (Ehrlich and Lacey 2013).
137,138) ZFP57/KAP1 complexes also interact with DNMTs and the hemimethylated DNA binding protein UHRF1 (also called NP95), which are essential for the maintenance of DNA methylation.
2007) UHRF1 plays a role in maintaining DNA methylation in mammalian cells.
Induction of apoptosis by thymoquinone in lymphoblastic leukemia Jurkat cells is mediated by a p73-dependent pathway which targets the epigenetic integrator UHRF1.
