TRPV3

A gene on chromosome 17p13.2 that encodes a calcium-permeable, non-selective cation channel that functions in a variety of processes, including temperature sensation and vasoregulation. The channel is expressed in subsets of sensory neurons that terminate in the skin and are activated at temperatures between 22ºC and 40ºC. TRPV3 may associate with TRPV1 and modulate its activity.

In the lab, the team produced one of the mammoth temperature sensors, a protein called TRPV3.
Reduced activity of TRPV3 may have led to similar temperature tolerance and physical changes in woolly mammoths.
The low pH strongly activates the TRPV3 ion channel, opening it and allowing calcium ions to flow into the cell.
Our experiments are the first to show that the TRPV3 ion channel is likely to be the target of the most effective skin enhancer in the cosmetics industry," said Jie Zheng, professor of physiology and membrane biology at UC Davis and one of the principal investigators of the study.
TRPV3 and TRPV4 dominant-negative (DN) isoforms were generated by isolating a truncated form of each channel, from 10 amino-acids N-terminal to the fifth transmembrane domain to 10 amino-acids C-terminal to the sixth ttansmembrane domain.

A second channel, TRPV3, responds to temperatures of about 85 degrees.
When TRPV3 was knocked out, the mice had a thinner outer skin layer with a dry, scaly texture, and appeared to be a less intact, more permeable barrier.
