SETD7

A gene on chromosome 4q28 that encodes a SET domain-containing histone-lysine methyltransferase that specifically monomethylates Lys-4 of histone H3, providing a specific tag for epigenetic transcriptional activation. SETD7 pays a central role in the transcriptional activation of genes encoding collagenase, insulin and other proteins; it also has methyltransferase activity toward non-histone proteins, such as p53/TP53, TAF10 and possibly TAF7. SETD7 is widely expressed, especially in pancreatic islets. It interacts with IPF1/PDX-1.

It was reported that renal mesangial cells induced the transcription of fibrotic genes in reply to TGF-[beta]1, and high glucose is due to the enrichment of active chromatin marks (H3K4me1/2/3, H3K9/14Ac) and the decrease of repressive markers (H3K9me2/me3) at promoters of these genes, along with the histone lysine acetyltransferase (p300/CBP) and histone lysine methyltransferase (SET7) occupancies at fibrotic gene promoters [62, 63].
In rat mesangial cells cultured in diabetic conditions or pretreated with TGF-[beta]1, expressions of a methyltransferase SET7 increased, along with the enrichment of SET7 at fibrotic gene Col1a1/Col4a1 promoters.

Of the enzymes that regulate this process, histone methyltransferases Set7 and Set9 are the most relevant, are activated in the hyperglycaemic settings, and mainly control genes involved in the inflammatory response [33].

Similarly, a study on human aortic endothelial cells acutely exposed to high glucose reported that the histone methyltransferase Set7 was mobilized to the p65 promoter, resulting in a significant increase in H3K4 monomethylation [22].
