A gene on chromosome 18q21.1 that encodes a basic helix-loop-helix (bHLH) transcription factor, which recognises an Ephrussi-box (E-box)-binding site first identified in immunoglobulin enhancers and expressed predominantly in pre-B-cells.

TCF4 mutations cause Pitt-Hopkins syndrome.
