KITLG

A gene on chromosome 12q22 that encodes KIT tyrosine kinase receptor ligand, which plays a key role in regulating cellular activities (such as: cell survival and proliferation; haematopoiesis; stem cell maintenance; gametogenesis; mast cell development; migration and function; and in melanogenesis) by activating multiple signalling pathways.

Molecular pathology
KITLG mutations cause familial progressive hyperpigmentation.

Slight decrease in Kitlg mRNA expression has also been observed in mice treated with busulfan, albeit this reduction was not statistically significant In contrast with GDNF and FGF2, KITLG is believed to be involved in SSCs differentiation.
Flence, it could be surmised that somatic cells composing SSCs niche respond to SSCs elimination by upregulation of GDNF and FGF2 and downregulation of KITLG in order to repopulate niche by germ cells.
This change affects how much KITLG is expressed in the hair follicles without changing how it's expressed in the rest of the body.
The change occurs over 350,000 nucleotides away from the KITLG gene and only alters the amount of gene expression about 20 percent - a relatively tiny blip on a biological scale more often assessed in terms of gene expression being 100 percent "on" or "off.
They found that KITLG activation by p53 is crucial for cancer processes, including cell proliferation, and found it was much more common in Caucasians than Africans, explaining why testicular cancer is more common among the latter.
Bell and Bond believe this variant may be selected in lighter-skinned men during evolution because the KITLG gene plays an important role in triggering the protective tanning response to UV rays.
The magnitude of risk linked to KITLG is much larger than that associated with genes affecting other cancers, such as breast, colon and prostate cancer.

However, since the prevalence of the common variant is so high, it may also reflect other underlying factors required in conjunction with KITLG for disease development.
