USP28A gene on chromosome 11q23 that encodes a deubiquitinase involved in DNA damage response, which interacts with isoform 1 of FBXW7; following DNA damage, it and dissociates from FBXW7 leading to degradation of MYC. USP28 is involved in DNA damage-induced apoptosis by deubiquitinating certain proteins of the DNA damage pathway (e.g., CLSPN). It is also acts at the G2 DNA damage checkpoint by deubiquitinating CLSPN and preventing its degradation by the anaphase-promoting complex/cyclosome (APC/C). It is downregulated by hypoxia.
