Each eluate sample (5 mL) from the MBD column was concentrated with Amicon Ultra Centrifugal Filter Devices (30 000 MWCO, Millipore) to 200 [micro]L and then extracted with phenol/ chloroform/ isoamyl alcohol (25: 24:1), precipitated in ethanol, and eluted in nuclease-free water.
Chloroform/ isoamyl alcohol (24:1) and phenol/chloroform/ isoamyl alcohol (25:24:1) extraction was performed consecutively and DNA was precipitated using 0.7 volumes isopropanol.
Then 50 uL of 3 M sodium acetate was added, followed by an equal volume of phenol: Chloroform: isoamyl alcohol (25:24:1).
The straight-chain higher alcohols are considered to have the most significant sensorial impact (1-propanol, 2-methyl-1-propanol (isobutyl alcohol), 2-methyl-1-butanol and 3-methyl-1-butanol (isoamyl alcohol).
Anhydrous iron(III) chloride (Fe[Cl.sub.3], Sd Fine Chemicals, India), ammonium hydroxide solution (30 w%, SRL, India), cyclohexane (SRL, India), isoamyl alcohol (SRL, India), and Triton X-100 (SRL, India) were of analytical grade and used as received.
The supernatant was extracted with 750 [micro]L of chloroform: isoamyl alcohol (24:1) by centrifugation at 15,000 rpm during 10 min to separate the phases.
This study investigates the effect of aging on the ability of Caenorhabditis elegans to chemotax from a point of origin to a known attractant, isoamyl alcohol. After measuring the chemotaxis index of Caenorhabditis elegans aged three days, five days and seven days, we analyzed results to determine whether or not a statistically significant difference in the chemotaxis indices was present.
