IFI6

IFI6

An (alpha- and beta-) interferon-inducible gene on chromosome 1p35 that encodes a protein which is thought to play a critical role in regulating apoptosis.

John Schoggins, Assistant Professor of Microbiology, used the cutting-edge CRISPR technology to identify the IFI6 gene as a potent antiviral gene targeting flaviviruses.
Ohlson performed the CRISPR screen that allowed us to uncover IFI6 as a potent suppressor of flavivirus infection," he said.
Schoggins explained that the CRISPR gene-editing technology made the study extremely efficient, uncovering the prominent flavivirus-inhibiting role of IFI6.
In cell culture studies, the IFI6 gene--apparently working via its protein product, also called IFI6 --inhibited yellow fever, a flavivirus known to infect the liver.
The mRNA levels of interferon-induced guanylate-binding protein 1 (GBP1), interferon alpha-inducible protein 6 (IFI6), beta-defensin 9 (GAL9), ICAM-1, HLAA, and BLB1 genes were significantly downregulated in the liver of assisted ducklings compared to that in normal ducklings.
The baseline expression levels of 25 IFN-regulated genes (MxA, GPR3, IL17RC, ISG15, TRAIL, OASL, IFIT1, IFIT2, RSAD2, OAS3, IFI44L, TRIM22, IL10, CXCL10, STAT1, OAS1, OAS2, IFNAR1, IFNAR2, IFN[beta], ISG20, IFI6, PKR, IRF7, and USP18) were examined but were not confirmed to have predictive value [53].
In addition to NLR inflammasomes (NLRP1, NLRP3, NLRP6, NLRP12, and NLRC4), there are four additional inflammasomes (AIM2, RIG-I, IFI6, and PYRIN) which form by non-NLR sensor proteins [57].
IFNL4, in turn, displays antiviral activities which are comparable to IFNL3 as shown in reporter cells expressing the IFNLR and a luciferase gene under the control of the IFI6 promoter [3].
