We reported that HYAL1 is overexpressed in PDAC cell lines and tissues and that inhibition of HYAL activity significantly inhibits the migration of PDAC cells [5].
For example, overexpression of HYAL1, one of the major HYALs, results in increased cell proliferation, migration, invasion, and metastasis in prostate and breast cancer [14, 15].
Human CTSG ELISA kit and human HYAL1 ELISA kit (Cusabio Biotech, Wuhan, Hubei, China), TGF-[beta]1, and KNG1 (Abcam, Cambridge, MA, USA) were applied to determine the concentration of these proteins in each serum of validation set (27 HC and 27 EH individuals).
In addition, to further investigate the plausible biological processes in which these proteins might be involved, we used bioinformatics analysis and the availability of commercial kits and discovered alterations in the biological process of factor production and catabolism process in many key pathogenesis nodes, and 4 candidates (proteins CTSG, KNG1, HYAL1, and TGF-[beta]1) were involved in the process and were selected for further verification.
Up-regulated genes included heparan sulfate proteoglycan 2 (HSPG2), hyaluronoglucosaminidase 1 (HYAL1) and interleukin 7 (IL-7), and down-regulated genes included interleukin 6 (1L-6), and C-reactive protein (CRP), as well as cathepsin G (CTSG), tachykinin receptor 1 (TACR1), chemokine (C-X3-C motif) ligand 1(CX3CL1), frizzled homolog 1 (FZD1), and insulin-like growth factor 2 (IGF2).
Genes up-regulated in the AR treatment group included HSPG2 and HYAL1, proteins involved in extracellular matrix generation and remodeling (Xu et al.
