HSPA14

A gene on chromosome 10p13 that encodes a member of the heat shock protein 70 (HSP70) family which is a component of the ribosome-associated complex (RAC), a heterodimer composed of Hsp70/DnaK-type chaperone HSPA14 and Hsp40/DnaJ-type chaperone DNAJC2. The RAC binds to nascent polypeptides, maintaining them in a folding-competent state while DNAJC2 stimulates HSPA14’s ATPase activity.

The importance of HSPs in maintaining proteostasis within oocytes takes on added significance in light of evidence that oocytes obtained from aged mice exhibit a marked decrease in the gene expression of Hsp40s (Dnaja2, Dnaja4, Dnajb1, Dnajb6, Dnajb10, Dnajb11, Dnajc3, Dnajc5, Dnajc8, and Dnajc21), Hsp70s (Hspa1b, Hspa4, Hspa8, Hspa9a, Hspa14, and Hsp70-3), Hsp90s (Hspcb), and components of the chaperonin containing TCP1 complex (Cct1-3 and Cct5) (Table 1) [3, 97].
Interestingly, expression of FISPA4 and HSPA14, which encodes ER chaperone FISP70, was down-regulated, though a co-chaperone of HSP70 encoded by DNAJB9, was upregulated.
