GRB10A gene on chromosome 7p12.2 that encodes an adapter protein which binds to, and suppresses signals from, activated receptors tyrosine kinases—e.g., insulin and insulin-like growth factor receptors (INSR and IGF1R, respectively)—by either interfering with the signalling pathway or increasing receptor degradation. It also downregulates Wnt signalling by interfering with the binding of AXIN1 to LRP6, and it may inhibit NEDD4-mediated degradation of KDR/VEGFR-2. GRB10 upregulates the KDR/VEGFR-2 signalling pathway.
