A gene on chromosome 17p13.1 that encodes a member of the Dishevelled (Dsh) protein family which undergoes post-translational phosphorylation and may play a role in the Wnt signal transduction pathway.

Using FDR p < 0.05, six enriched pathways were identified including "Notch Signaling Pathway" (genes NOTCH4 and DVL2), "Rlio GTPase cycle" {FAM 13A; HMHA1; VAV2; and GMIP), "Neurotransmitter Release Cycle" (HSPA8; and RIMS1), and "GABA synthesis, release, reuptake and degradation" (HSPA8; and RIMS1).
From more than 12 million individual differences, they identified one mutation, in a gene called "Dishevelled 2," or DVL2, that was found in all the bulldogs and French bulldogs sampled.

Therefore, cir-ITCH likely plays an inhibitory role in ESCC and colorectal and lung cancer, through promoting ITCH-mediated ubiquitination and subsequent proteasome-mediated degradation of phosphorylated Dvl2 scaffold protein, which impairs the canonical Wnt/[beta]-catenin signaling [104-106].
