CLEC7A

A gene on chromosome 12p13.2 that encodes a C-type lectin domain protein which acts as a pattern receptor for beta-1,3-linked and beta-1,6-linked glucans found in pathogenic bacteria and fungi. CLEC7A is required for the TLR2-mediated inflammatory response and for TLR2-mediated activation of NF-kappB. It enhances cytokine production in macrophages and dendritic cells, plays a role in T-cell activation and proliferation, and mediates production of reactive oxygen species and phagocytosis of Candida albicans.

Eng1 acts trimming [beta]-glucan segments exposed on the fungal cell surface further minimizing potential Dectin1 recognition, decreasing production of proinflammatory cytokines by phagocytes thereby enhancing Histoplasma ability to escape detection by host phagocytes.
However, the increased levels of dectin1 mRNA expression were marginal ([DELTA][DELTA]CT = 0.76 [+ or -] 0.20) and not comparable to those following LPS stimulation in cultures ([DELTA][DELTA]CT = 10.90 [+ or -] 3.61).
In contrast, challenge with LPS resulted in downregulation of dectin1 expression in GM-CSF/IL-4 differentiated human PBMC (data not shown), which agrees with previous studies [30,31].
