CCNT1

A gene on chromosome 12q13.11 that encodes a member of the highly conserved cyclin family, which regulates CDK kinases. Cyclin T1 binds tightly to CDK9, forming a major subunit of the transcription elongation factor p-TEFb, which interacts with, and acts as a cofactor of, HIV-1’s Tat protein, and is required for full activation of viral transcription.

Provision of mouse cells with human CD4, CCR5 and cyclin TI (cycT1) has uncovered a block to HIV assembly or release.
An HIV-based vector encoding cycT1 was introduced into the entire collection.
Initial work has involved the introduction of the cDNA library into mouse cells containing cycT1. The cDNA and vector were rescued by transfecting the mouse cells with packaging vector and vesicular stomatitis virus G (VSV G).
