First, the deletion was concerned with an in-frame deletion of 5 amino acids within a highly evolutionarily conserved region in the CL domain of Cx50 protein, which binds Ca [sup]2+/CaM to mediate gap junction channel in the lens of the eye.
These mutations occur at the NT, TM1 to TM4, E1 and E2, or CT,[sup][27] but none has been reported on the CL domain [Figure 1]d and Supplementary Table 4.
In this study, by combined NGS and targeted genomic enrichment technology, we identified a novel 15 deletion in a highly conserved region of CL domain of the GJA8 gene associated with cataracts in a three-generation Chinese family with ADCC (mutations have never reported in CL domain before).
