Preclinical studies suggest that Cdk5 is a gene that is important for the normal wiring of the brain during early development and may be involved in some neurodegenerative disorders, including ALS, Parkinsons and Alzheimers disease.
A[beta] activates NLRP3 inflammasomes via the P2X7R receptor or TLR4-MyD88-NF-[kappa]B pathway, [alpha]-synuclein activates the toll-like receptor 2 (TLR2) which promotes the assembly of NLRP3 and induces IL-1[beta] synthesis, ROS activates NLRP3 through c-Abl kinase or upregulates CTSB activity, Cdk5 could activate NLRP3 inflammasome, caspase-1 could cleave [alpha]-synuclein into a highly aggregation-prone specie, and NLRP10 attenuates A[beta]- induced caspase 1 activation and IL-1[beta] release.
Various important kinases regulate tau phosphorylation, including cyclin-dependent kinases (CDK2 and CDK5), GSK-3[beta], mitogen-activated protein kinase (MAPK), extracellular signal-regulated protein kinase 1/2 (ERK1/2), c-Jun N-terminal kinase (JNK), Akt, protein kinase a (PKA), and calcium-calmodulin protein kinase 2 (CaMKII).

Conversion of p35 to p25 deregulates Cdk5 activity and promotes neurodegeneration.
James Bibb, Associate Professor of Neurology and Neurotherapeutics at UT Southwestern said that if people inhibit Cdk5, then the vast majority of brain tissue stays alive without oxygen for up to one hour, which tells them that Cdk5 is a central player in nerve cell death.
GSK3, PKC, PKA, 14-3-3 Mode 1, PKG, p38MAPK, cdk5, CKII, cdc2, ATM, and CKI are the most predicted kinases involved in phosphorylation of many serine and threonine residues in IRS-2.

Creating highly selective CDK2 compounds is a challenge due to the identity of ATP binding subsites within CDK1, CDK2, and CDK3 molecules; CDK2 also possesses 92% and 80% sequence identity in CDK5 and CDK6 molecules, respectively (RCSB Protein Data Bank code: 1b38).
Interestingly, cyclin-dependent kinase 5 (Cdk5) was reported to mediate phosphorylation at Ser56 of vimentin [46].
