The expression of CD16 on NK cells can undergo considerable down-regulation in cancer patients, which significantly inhibits the immune system's antitumor response.
Three monocytes subpopulations, based on their expression of CD14 and CD16 surface markers, have been described in humans [13].
Granulocytes were gated according to their morphological characteristics and then analyzed for the expression of CD15, CD16, and CD11b.
The abnormal NK cells were positive for surface CD45 (high), CD2, CD26, CD38, CD94 (high), and HLA-DR and cytoplasmic granzyme B and perforin; and they were negative for surface CD3, CD4, CD5, CD7, CD8, CD11c, CD16, CD19, CD20, CD25, CD56, Ig kappa, and lambda chains.

In addition, it was also found that the sub-population of CD16 and CD14 cells and DNases associated with monocytes increased in patients with some autoimmune diseases (grave disease, multiple sclerosis).

Mature monocytes expressing CD16 showed higher levels of genes and corresponding proteins associated with phagocytic activity mediated through Fc[gamma] receptor: actin-related protein 2/3 complex (ARPC5), CD16, hemopoietic cell kinase (HCK), thyrosine-protein kinase Lyn (LYN), hemeoxygenase 1 (HMOX), villin 2 (VIL2).
The flow cytometric study of the peripheral blood showed that the circulating neoplastic cells (comprising about 40% of total white blood cells) seemed to be of NK-cell lineage with expression of CD45 (bright), CD2, CD56, CD11b, CD16 (partial), and cytoplasmic CD3, but the sample was negative for surface CD3, CD57, CD4, CD5, CD7, CD8, TCR, CD123, terminal deoxynucleotidyl transferase, CD34, B-cell markers, or myeloid markers (Figure 2, A through F).
Anti-HNA-2a and anti-Fc[gamma]RIIIb (also known as CD16) in mothers with an HNA-null phenotype [i.e., FcRyOIb deficiency (Box 1)] are more rarely involved (5).
Blood granulocytes from some of the individuals with PIGV mutations had partially reduced surface expression of CD16, a GPI-AP.
Depending on the affinity with which immunoglobulins bind to the receptor, the FcyR receptors are classified either as high-affinity --known as FcyRI (CD64)--or as low-affinity receptors, such as FcyRII (CD32) and FcyRIII (CD16).
