DIAPH3A gene on chromosome 13q21.2 that encodes a homolog of the Drosophila diaphanous gene which binds to GTP-bound Rho and profilin, acting in a Rho-dependent manner to recruit profilin to the membrane, where it promotes actin polymerisation. DIAPH3 is required for cytokinesis, stress fibre formation and transcriptional activation of serum response factor. DIAPH proteins couple Rho and Src tyrosine kinase during signalling and regulation of actin dynamics.
