ALDH2

A gene on chromosome 12q24.2, which encodes a protein belonging to the aldehyde dehydrogenase (ALDH) family of proteins, and is the second enzyme of the major oxidative pathway of alcohol metabolism. The two major liver isoforms of ALDH, cytosolic and mitochondrial, can be distinguished by their electrophoretic mobilities. Most Caucasians have both; 50% of Asians have only the cytosolic isozym, making them more susceptible to acute alcohol intoxication.

Therefore, we investigated whether the expressions of Bax and Bcl-2 genes are related to ALDH2 induced protection in diabetic myocardial I/R injury.
This finding indicates that ADH 1B*2 may be protective against alcohol dependence even in the presence of the fully active ALDH2 allele, suggesting even mild discomfort experienced because of alterations in acetaldehyde metabolism may prevent overindulgence in alcohol.
Although the studies described above demonstrate that ADH1B and ALDH2 variants influence the risk of AUD, it also is clear these genes and their alleles do not act in isolation.
The severe effects of the ALDH2*2 variant on acetaldehyde levels and alcoholism risk demonstrate that the mitochondrial ALDH2 enzyme normally is the most important enzyme for eliminating acetaldehyde from the body and keeping acetaldehyde levels extremely low.
Single nucleotide polymorphisms of ADH1B, ADH1C and ALDH2 genes and esophageal cancer: A population-based case-control study in China.
One of the best understood polymorphisms of alcohol-metabolizing enzymes is associated with the ALDH2 enzyme.
As noted previously, a deficiency of the ALDH2 enzyme in people carrying the ALDH2 Lys487allele contributes to an elevated risk of cancer from alcohol consumption.
The purpose of this article is to review genotype (1) prevalence rates of three genes, the aldehyde dehydrogenase gene ALDH2 and the alcohol dehydrogenase genes ADH1B and ADHIC.
An ALDH2 variant shown to have protective effects primarily in East Asians was not found in either Trinidadian ethnic group.
