In its first phase, phosphohydrolase hMTH1 (human 8-oxo-dGTPase --a homologue of the bacterial enzymatic protein MutT) removes the defective nucleotide, thus preventing its incorporation into DNA, and then glycosidase hydrolyzes the N-glycosidic bond between the modified base and 2'-deoxyribose.
Inhibition of 8-oxo-2'-deoxyguanosine 5'-triphosphate pyro-phosphohydrolase (8-oxo-dGTPase) activity of the antimutagenic human MTH1 protein by nucleoside 5'-diphosphates.
