Cells were cytocentrifuged (Cytospin 3; Shandon Scientific Ltd, Grupo Taper, Madrid, Spain) and cell smears were allowed to air dry and were stained for 15 minutes at room temperature. Cytocentrifuged slides of spun-down lavaged cells were prepared for each animal and stained with Diff-Quik (Dade Behring, Newark, DE, USA) for cell differential counts. Then, the PECs were cytocentrifuged (500 rpm for 5 minutes) onto microscope slides and permeabilized in 100% methanol for 3 minutes. |
