NOS2

A gene on chromosome 17q11.2-q12 that encodes inducible nitric oxide synthase (iNOS), a small molecule that has various roles in cellular functions and acts via a cGMP-mediated signal transduction pathway. iNOS is both tumouricidal and bactericidal, has nitrosylase activity, and mediates cysteine S-nitrosylation of cytoplasmic target proteins such COX2. It is regulated by calcium/calmodulin, induced by endotoxins and cytokines, including IFN-gamma, in synergy with bacterial lipopolysaccharides (LPS, TNF or IL1B/IL-1 beta). Aspirin inhibits its expression and function. Some genetic variants of NOS2 play a role in resistance to malaria. NOS2 is also a less preferred gene symbol for what is now designated as NANOS2, see there.

After the cells were stimulated by Hx for different time, the gene expressions of COX-2, PGIS, iNOS, eNOS, HIF-1a, HIF-2a, and VEGF were detected by real-time RT-PCR [Figure 1].

Inducible nitric oxide synthase (iNOS) catalyzes the oxidative deamination of L-arginine to produce NO.
In the present communication, we wish to report the inhibitions of NO production of lipopolysaccharide (LPS)-stimulated RAW 264.7 cells by seven herbertenoids (3-7, 10, 11) and two cuparenoids (8, 9) isolated from liverworts, as well as the inhibition of LPS-induced inducible nitric oxide synthase (iNOS) by the representative compounds (9).

In a recent preliminary study following exposure to hypergravity (20), we observed a significant elevation of iNOS mRNA expression levels in the livers of mice, suggesting that exposure to hypergravity is a biophysical condition that can adversely affect the liver.
It is suggested that the cells which are present from a significant distance from iNOS source may undergo apoptosis, while cells close to iNOS source may degrade the p53 protein formed resulting in protection from apoptosis, which suggest a special relationship in cells with regard to the effect of NO on apoptosis [31,32].
