The advantage of Sanger sequencing is the ability to detect any mutation within the PCR amplification product without prior knowledge of the location or the type of mutation; however, Sanger sequencing is limited by a low
analytic sensitivity of 15% to 20%, which limits its ability to analyze samples with low tumor cellularity.
