Bound antibody can be recovered by acid elution (14).
A potential drawback of affinity purification is that the obtained DAT could be denatured by acid elution. This risk could be minimized by immediately neutralizing pH by adding 1 mol/L Tris, followed by dialysis with phosphate-buffered saline.
Results are consistent with the hypothesis that HLA-A2 antigens were adsorbed from the platelets' surface by
acid elution. They also elucidate the difference in the sensitivity of CDC and ELISA methods to detect antibodies.
Scientists then remove the adsorbents from the water and the metals are readily extracted using a simple
acid elution method.
(2) We found that the Rosette and acid elution techniques were effective at detecting a fetomaternal hemorrhage (FMH) requiring additional units of Rh immune globulin (Rhlg; sensitivity 1.0 and 0.96, specificity 0.75 and 0.92, respectively).
In our view, improving the effectiveness of RhIg dosing would require 2 things: (1) a more accurate calculation of maternal blood volume, such as Dr Ramsey's RhIg Dose Calculator2; and (2) replacement of acid elution FMH quantitation with flow cytometry in all cases in which additional Rh immune globulin doses are indicated by initial Rosette or acid elution test results.
Actually, the fetal cells are resistant to this
acid elution and stain a bright pink with the method we use.
The [[alpha].sub.2]-MG affinity-chromatography
acid elution pattern is shown in Fig.
The following solutions were prepared for the extraction and spotting of basic drugs from urine according to manufacturer's recommendations: 1.0 mol/L acetic acid, wash reagent I (0.1 mol/L acetic acid), wash reagent II (100 mL of ethyl acetate, 100 [micro]L of glacial acetic acid),
acid elution reagent (140 mL of hexane, 60 mL of ethyl acetate, 200 [micro]L of glacial acetic acid), and basic elution reagent (98 mL of ethyl acetate, 2 mL of concentrated ammonium hydroxide).
We are planning to have these employees perform
acid elution fetal cell counts and bone marrow differentials in the near future.
