In this study, our finding showed that the aged ApoE-/- mice had lower endothelium-dependent vasorelaxation elicited by acetylcholine in aortic segments mounted in myographs. In contrast, TXL improved endothelium-dependent vasodilatation in ApoE-/- mice, indicating its protective effect on the vascular ECs, which proved the results of other clinic trials,[13] and that mechanism probably correlated with elevating eNOS activity and increasing NO synthesis.[14]
After sacrifice, aorta was immediately isolated and mounted onto myograph. (A) Aortic contractile response to phenylephrine (PE).
Maximal voluntary isometric force was measured using a myograph which consisted of a load cell (100 N), a peak-and-hold meter and a visual feedback monitor.
Mesenteric arteries were dissected free from surrounding tissue and mounted on a small vessel wire myograph for measurement of isometric tension (n=5/gender/group).
Myograph Family Mortuary of Eugene is in charge of arrangements.
The heart was dissected free, after removal of chest plate, and connected by means of a hook to a "B" myograph. Impulses from the myograph were fed into an E and M physiograph and recorded on a moving paper.
Uterine horn contractions were monitored on a myograph (Model F-60, Narco Bio-Systems) connected to a pen recorder (Narcotrace 40 physiograph, Narco Bio-Systems) set at a sensitivity of 50 mV/cm.
Vasodilatation was measured in mesenteric arteries with a wire myograph. Nitrites were measured by fluorescence with 2,3-diaminonaphthalene and [H.sub.2][O.sub.2] was measured with carbon microsensors.
