Epicotyl sections as targets for plant regeneration and transient transformation of common bean using Agrobacterium tumefaciens.
The
epicotyl (Figure 1D) and hypocotyl (Figure 1E) segments obtained from in vitro germinated seedlings (Figure 1C) that were segmented and inoculated in MS medium (Experiment I), and WPM medium supplemented with 2.0 mg.[L.sup.1] of BAP (Experiment II), and both media added with 30 g[L.sup.1] of sucrose, 4.0 g[L.sup.-1] of agar, with different doses and combinations of growth regulators.
Seedlings (Figure 1A and C) are phanerocotylar and epigeal, with axial root, collet visible for presenting slight constriction, hypocotyl long, cylindrical and green, and
epicotyl relatively short, cylindrical, green, hairy, with scale like leaves.
After three weeks, the seedlings were excised and separated into two portions: hypocotyl segment including the cotyledonary nodes and
epicotyl segment with the apex.
Seedlings obtained from cytokinin treatments (24 mg L-1 BA) were stronger with thick hypocotyls, reduced
epicotyls and primary roots with few or no lateral root formation.

durum in filter paper soaked with cell-free extract containing MC-LR at concentrations between 1,600-11,600 mg [L.sup.-1], a dose-dependent relationship was observed in both germination inhibition and length decrease of the
epicotyl and primary root as well as in the number of lateral roots due to abnormalities caused to the pericycle.
The seed was considered germinated by a visible protrusion of split seed coat with the cotyledons, hypocotyls, and
epicotyl on the surface of the soil.
