CLEC7A

A gene on chromosome 12p13.2 that encodes a C-type lectin domain protein which acts as a pattern receptor for beta-1,3-linked and beta-1,6-linked glucans found in pathogenic bacteria and fungi. CLEC7A is required for the TLR2-mediated inflammatory response and for TLR2-mediated activation of NF-kappB. It enhances cytokine production in macrophages and dendritic cells, plays a role in T-cell activation and proliferation, and mediates production of reactive oxygen species and phagocytosis of Candida albicans.

Recently, several studies have reported that Syk is a key molecule in the signaling pathways initiated by pattern recognition receptors (PRRs) in terms of recognizing pathogen-associated molecule patterns (PAMPs) and activating innate immune responses mediated by Syk is activated through the interaction with C-type lectin CLEC7A, which is a mammalian PRR for fungal [beta]-glucans [68], and this Syk-CLEC7A interaction stimulates its signaling pathways, resulting in the production of ROS [69, 70] and phagocytosis of yeast by dendritic cells [68].
Dectin-1 (dendritic cell-associated C-type lectin-1, DCAL-1, Clec7A) or beta-glucan receptor is a C-type lectin receptor which is part of the NK gene complex in the Dectin-1 cluster (Table 1 and Figure 1) [134].
