This is important because previous studies focused on blocking the NMDA receptors in order to prevent cell death, but this approach didn't work because it affected normal cell function and had a relatively short therapeutic window.
However, the use of thrombolytics (tPA) is limited due to the therapeutic window of < 3-6 hours post onset of stroke symptoms such that only a small fraction of stroke patients receive this therapy.
On account of significant enterohepatic recirculation, and the fact that MPA excretion does not follow first-order kinetics, trough plasma MPA levels are inadequate to establish a therapeutic window of treatment efficacy and toxicity.
