mitoxantrone /mi·to·xan·trone/ (mi″to-zan´trōn) a DNA-intercalating anthracenedione, used as the hydrochloride salt as an antineoplastic agent in the treatment of acute myelogenous leukemia and advanced, hormone-refractory prostate cancer; also used in the treatment of secondary multiple sclerosis.

indications It is prescribed in combination with other approved drugs in the initial treatment of acute nonlymphocytic leukemia in adults and has activity against other leukemias, lymphoma, and breast cancer.

contraindications It should not be given to patients who are pregnant, have a clinically significant low left ventricular ejection fraction, or have evidence of prior hypersensitivity to this drug. Its use should be accompanied by close and frequent monitoring of hematologic and chemical laboratory parameters, as well as frequent patient observation.

adverse effects The side effects most often reported include hypotension, urticaria, dyspnea, rashes, phlebitis, myelosuppression, nausea, and vomiting.

It is also used to reduce the frequency and severity of relapses in patients with secondary multiple sclerosis.
