Potent and selective inhibition of nitric oxide-sensitive guanylyl cyclase by 1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one. The paper elucidates the means by which nitric oxide and carbon monoxide bind to a protein called soluble guanylyl cyclase (sGC) which is critical in the formation of another protein, cGMP, which induces the relaxation of blood vessels. Nitric oxide is a biogenic messenger, an EDRF (15,16), and it activates the guanylyl cyclase system (17) [it converts guanosine triphosphate to cyclic guanosine monophosphate (cGMP)], raising the cGMP pool and therefore inducing, among other things, vasodilatation (17) by lowering the intracellular calcium ion (16). |
