dihydrofolate reductase
enzyme catalyzing the conversion of folate to 5,6,7,8-tetrahydrofolate, which is the key carrier of one-carbon units in purine and pyridime synthesis, the pathway for the breakdown of histidine and the synthesis of S-adenosylmethionine from S-adenosylhomocysteine. This enzyme is the target for methotrexate, which inhibits enzyme action, a key means of inducing remissions in acute leukemias.

Recent work demonstrated that EGCG reduced dihydrofolate reductase activity, which would affect nucleic acid and protein synthesis.
Pemetrexed is an antifolate/antimetabolite with several molecular targets involved in both pyrimidine and purine neosynthesis: thymidylate synthase (TS), dihydrofolate reductase (DHFR) and glycinamide ribonucleotide formyltransferase (GARFT).
Dihydrofolate reductase is required for the growth of actively dividing cells.
