| Each positive PCR product was subjected to DNA sequencing with fluorescent-labeled dideoxynucleotide technology (BigDye Terminator Cycle Sequencing Ready Reaction Kit; Applied Biosystems, Foster City, CA, USA). Nucleotide sequences were determined by oligonucleotide-directed dideoxynucleotide chain-termination sequencing reactions utilizing 300 ng of template DNA, forward (M13F) and reverse (M-13R) primers, fluorescently-labeled dideoxynucleotides and AmpliTaq FS DNA polymerase in a cycling sequencing method. Other approaches to improving sequencing technology involve methods that are independent of the Sanger dideoxynucleotide chain termination reaction or of electrophoretic separation of the termination products. |
