To further reduce this risk, uracil-N-glycosylase and deoxyuridine triphosphate were used to prevent amplicon carryover (13). Briefly, 1 [micro]g of sample RNA and global reference RNA (Stratagene, La Jolla, CA) were converted to cDNA with reverse transcriptase and then amplified using T7 RNA polymerase while labeling with either cyanine 3 deoxyuridine triphosphate (Cy3-dUTP) or Cy5-dUTP (Low RNA Input Linear Amplification Kit, Agilent Techologies). Enhanced breadth of priming is achieved through the use of primers and probes containing 5-propynyl deoxycytidine and deoxyuridine nucleotides that offer increased affinity and base pairing selectivity (18,19). |
