DUSP1

DUSP1

A gene on chromosome 5q34 that encodes dual-specificity phosphatase 1, a protein which dephosphorylates MAP kinase MAPK1/ERK2. DUSP1 downregulates cell proliferation and is thought to play a key role in cellular responses to environmental stress. It is also a less preferred gene symbol for what is now designated as DUSP12, see there.

The stimulation of DUSP1 expression by GA and 18[beta]GA occurred via both glucocorticoid receptor (GR) and PI3K signaling, and the simultaneous activation of transcription elements, such as AP1 (activator protein 1), CRE (cAMP response element), GRE (glucocorticoid receptor element) and NFAT (nuclear factor of activated T-cells), was confirmed.
The glucocorticoid receptor can reduce MAPK signaling by up-regulating dual specificity protein phosphatase 1 (DUSP1) expression, but DUSP1 is also regulated by MAPK.
The following primers were employed to amplify DUSP1 and the internal control RPL13A: DUSP1, forward = 5'-AGGCAGGCATETCCCGGTCA-3', reverse = 5' -TGCAGCAGCTGGCCCATGAA-3'; RPL13A, forward = 5'-CCGGAAGAAGAAACAGCTCATGAGG-3', reverse = 5'-GCAGGCA-ACGCATGAGGAATTAAC-3'.

This study employed DUSP1 mRNA expression as evidence of GR activation.
Previous studies from our laboratory have shown both DUSP1 (dual specificity phosphatase 1) and IL8 (interleukin 8) to be robust biomarkers for detection of oral squamous cell cancer (2, 7, 24, 25).
