DAXX

A gene on chromosome 6p21.3 that encodes a multifunctional protein located in the cytoplasm and nucleus which, as an adaptor protein in the MDM2-DAXX-USP7 complex, regulates RING-finger E3 ligase ubiquitination activity. It is thought to mediate activation of the JNK pathway and apoptosis via MAP3K5, in response to signalling from TNFRSF6 and TGFBR2.

DAXX protein was related to FAS protein which belongs to the tumor necrosis factor receptor superfamily and its intracellular tail contains death domain which is critical for apoptotic signaling.
Loss of DAXX and ATRX are associated with chromosome instability and reduced survival of patients with pancreatic neuroendocrine tumors.
Two other commonly mutated genes, DAXX and ATRX, which had not previously been linked to cancer, also have epigenetic effects on how DNA is read.
ATRX, DAXX and MEN1 mutations have better prognosis.
