Then, the cells were stained with 1 [micro]g/ml DAP1 solution for 5 min at room temperature in the dark.
Cover slips were carefully washed with PBS, fixed with 4% paraformaldehyde for 10 min and incubated with 10 [micro]g/ml DAP1 for 10 min.
Then, the slides were triple stained with p-VEGFR2 (FLK1/KDR) primary antibody (Cell Signaling, 3817; 1:100), antirabbit IgG conjugated with Alexa Fluor 555 (Cell Signaling, 4413; 1:500) and ProLong Gold antifade reagent with DAP1 (Invitrogen).
Only living cells, identified by DAP1 to stain dead cells were considered for the analyses.
Nuclei were counterstained with DAP1 (4,6-diamidin-2-phenylindol) (1 [micro]/ml) or propidium iodide (1 [micro]/ml).
The slides were afterwards mounted in Prolong Gold anti-fade reagent with DAP1 (invitrogen Molecular Probes), and were inspected with a confocal microscopy (Zeiss 710).
The first was with the DAP1 filter to detect the blue-white fluorescence of calcofluor binding mainly to chitin in the fungal cell wall.
After washes in PBS, slides were incubated with a fluorescein-isothiocyanate-conjugated (FITC) anti-mouse second antibody (1:50; DingGuo, China) and 5 [micro]g/ml DAP1 for 40 min at room temperature.
