Stimulation was performed with 40 [micro]L/mL (total protein content of approximately 1 [micro]g/mL) of vaccinia viral antigen resuspended according to the manufacturer's instructions (Maine Biotechnology Services, Portland, ME), or 2 [micro]g of CMV antigen (Biowhittaker, Walkersville, MD), always in the presence of co-stimulation with both anti-CD28 and CD49d monoclonal antibodies (1 [micro]g/mL, Becton, Dickinson and Company, Franklin Lakes, N J). Levitsky's laboratory research involves basic studies of antigen processing and presentation, T-cell co-stimulation, T-cell priming versus tolerance, and the evolution of tumor-specific immunity during immune reconstitution. Abatacept is the first of a class of drugs, called co-stimulation blockers, that selectively impede one of the two signals needed to activate T cells. |
