amastigote /amas·ti·gote/ (ah-mas´tĭ-gōt) the nonflagellate, intracellular, morphologic stage in the development of certain hemoflagellates, resembling the typical adult form of Leishmania.

amastigote
an early stage in the life cycle of trypanosomes, found in invertebrates. The body is rounded and the flagellum is absent or represented by a short fibril. The kinetoplast is present.

The central scale was removed from one of the lesions, and scrapings from the base of the lesion were stained with Giemsa stain, which showed intracellular amastigotes with peripheral nuclei and rod-shaped kinetoplasts.
Ideally, scrapings from underneath the undermined lesion's edge reveal amastigotes, but a wealth of confirmatory indirect methods exists.
