ADME parameters

The critical pharmacokinetic hurdles —absorption, distribution, metabolism, elimination—that a drug must address before being approved by the FDA for clinical use. Of the four, metabolism (biotransformation) is regarded as the most important and complex, as it affects oral bioavailability, systemic exposure, inter-subject variability and drug-drug interactions.
