Briefly, aorta cryosections were incubated with a dye quenched-gelatin (EnzChek; Molecular Probes, Invitrogen, Carlsbad, CA) and 1 [micro]g/mL 4',6-diamidino-2-phenylindole (DAPI; nuclei stain, Invitrogen) for 6 hr in a dark, humid chamber at 37[degrees]C.
After another phosphate buffered saline wash, cells were incubated with 4',6-diamidino-2-phenylindole (Invitrogen) for 10 minutes in the dark.
The nuclei of the cells were counterstained with 1 mg/L 4',6-diamidino-2-phenylindole.
All coverslips were mounted with mounting medium containing a proper dilution of 4',6-diamidino-2-phenylindole (DAP1) to counterstain DNA in the nuclei.
The slides were washed 3 times in 4X SSC containing 2 mL/L Tween 20, counterstained with 4',6-diamidino-2-phenylindole dihydrochloride (Sigma-Aldrich) to allow for chromosome identification, and mounted in phenylenediamine mounting medium.
